QUANTIFERON TB GOLD IN-TUBE
Specimen Collection Requirements
1 mL patient blood in each of three specialized blood collection tubes: Nil control tube (grey top), Mitogen control tube (purple top), and TB antigen tube (red top). All three tubes and instructions are in the QFT Single Patient Pack, Part #116768. Contact any SHS laboratory for supplies.
Immediately after collection, shake tubes vigorously up and down for 5 seconds (or 10 times) to ensure the entire inner surface of the tube is coated with blood (proper mixing is needed to allow t-cells to contact antigen). Frothing is expected. If tubes cannot be incubated at the collection site, label all tubes as “NOT INCUBATED.” NOTE: It is critical that incubation of the tubes occur at the performing lab within 16 hours of collection. Do not refrigerated or freeze.
If tubes cannot reach the performing lab within 16 hours, they must be incubated at the collection site. Within 16 hours of collection, shake the tubes vigorously up and down for 5 seconds (or 10 times) and incubate UPRIGHT at 37° C for 16-24 hours. After incubation, centrifuge the tube for 15 minutes at 2000-3000 RCF (g) prior to transport to the Central Laboratory. Incubated specimens must be transported at 2-8° C. Label all tubes as “INCUBATED”.

NON-INCUBATED TUBES: Ambient, tubes must reach testing lab within 16 hours of collection.
Frozen specimen (both INCUBATED and NON-INCUBATED tubes); specimen not incubated within 16 hours of collection; specimen centrifuged prior to incubation; refrigerated specimen (NON-INCUBATED tubes).
INCUBATED TUBES: 2 hours ambient or 28 days refrigerated. Unstable frozen.
NON-INCUBATED TUBES: 16 hours ambient. Unstable refrigerated or frozen.
